Effect of NSAIDs on Bone Healing Rates: A Meta-analysis.

Summary

This meta-analysis systematically reviewed the literature to determine whether NSAID use leads to delayed union or nonunion of bones. The analysis found that NSAID exposure significantly increased the risk of delayed union or nonunion, with an odds ratio of 2.07, meaning patients taking NSAIDs were approximately twice as likely to experience impaired bone healing compared to those not taking NSAIDs. Importantly, the study found that this effect was dose and time dependent. Low-dose, short-duration NSAID exposure did not significantly affect bone union rates, with an odds ratio of 1.68 that was not statistically significant. In pediatric patients, NSAIDs did not have a significant effect on bone healing. These findings demonstrate that while brief, low-dose NSAID use may be safe for bone healing, extended or high-dose use significantly impairs the body's ability to heal bone. This supports the recommendation that NSAIDs should be used at the lowest effective dose for the shortest duration possible, and that patients with fractures or recovering from bone surgery should be particularly cautious. The dose-dependent nature of the risk reinforces that timing and dosing strategy matter significantly when using these medications during recovery.
