The review found that at low doses of 800 to 1,200 mg per day, which are the doses approved for over-the-counter use in many countries, ibuprofen has a safety profile comparable to paracetamol (acetaminophen). Its pain-relieving activity is directly linked to its anti-inflammatory effects, specifically through the reduction of cyclooxygenase (COX-1 and COX-2) enzymes that produce inflammatory prostaglandins. Unlike paracetamol, ibuprofen does not carry a risk of liver damage, and unlike aspirin, it does not cause irreversible changes to body tissues. The review highlights that ibuprofen has a short plasma half-life, meaning it clears the body relatively quickly, which contributes to its low toxicity compared to other NSAIDs that remain in the body longer.
